P40是核转录因子P63的一种亚型，其可通过抑制细胞分化基因的表达来使细胞维持在具有再生潜能的未分化状态，参与组织的发育和修复。P40在标记鳞状上皮时具有较高的特异性，且与P63不同的是其在腺癌和淋巴瘤中不存在交叉反应。因此，P40已成为临床上诊断头颈部鳞癌、食道鳞癌、宫颈鳞癌等鳞状细胞癌的主要标志物之一，且其表达水平与癌症的恶性程度呈正相关关系，这可能是由于P40通过促进上皮-间质转化(EMT)来参与肿瘤发展。其中，P40与肺腺癌标志物的联合应用对肺鳞癌的鉴别诊断发挥了重要的指导作用。但有学者发现，在部分非小细胞肺癌中存在P40和用以标记肺腺癌的TTF-1的共同表达，这为肺部肿瘤的发病机制提供了一个新的研究方向。
