Signal transducer and activator of transcription 6 (STAT6) is a core factor in the IL-4 and IL-13 signaling pathways. It can be catalyzed by the Janus kinase (JAK) to phosphorylate and form dimers, which will bind to specific DNA sequences and regulate transcription to participate in immune regulation and cell proliferation. In clinical practice, STAT6 is a highly specific and sensitive diagnostic biomarker for solitary fibrous tumor (SFT). This is because the gene fusion of NAB2 and STAT6 is commonly present in SFT, which will not only lead to abnormal expression of STAT6 in the nucleus, but also invalidate the inhibitory effect on early growth response 1 (EGR1) mediated cell proliferation signal, resulting in cellular carcinogenesis. In addition, some research has found that STAT6 is expressed in various tumors such as liver cancer, gastric cancer, colorectal cancer, prostate cancer, and breast cancer. It can promote the proliferation and metastasis of cancer cells by continuously activating JAK-STAT6 signaling pathway, which is closely related to the occurrence and development of tumors.
