Polo like kinase-1 (PLK-1) is a highly conserved serine/ threonine protein kinase that widely exists in eukaryotic cells. It is one of the important cell cycle regulatory factors, which plays a crucial role in mitosis activation and DNA damage repairing. However, some research has found that PLK-1 is abnormally overexpressed in lung cancer, kidney cancer, gastric cancer, colorectal cancer, breast cancer, lymphoma, melanoma, glioma and other malignant tumors, which may be because the high expression of PLK-1 will cause the mitosis malfunction and promote the cell disordered proliferation, thereby inducing the malignant transformation of cells and enhancing the invasion ability of tumors. Therefore, in clinical practice, PLK-1 can serve as an important biomarker for evaluating the degree of cancer proliferation and infiltration, and the abnormal increase in its expression level is closely related to the poor prognosis of patients. In addition, PLK-1 has become a new research highlight in tumor targeted therapy.
