MOC-31 is a monoclonal antibody initially targeting small cell lung cancer cells, which can specifically recognize and bind to the first epithelial growth factor like sequence of epithelial cell adhesion molecule (Ep-CAM/Ber-EP4). Some research suggested that Ber-EP4 is highly expressed in epithelial tumors such as lung cancer, gastric cancer, colorectal cancer, and ovarian cancer, but not or lowly expressed in mesothelium and its derived tumors. Therefore, MOC-31 is mainly used in the diagnosis of epithelial tumors, especially as a differential diagnostic marker between adenocarcinoma (+) and malignant mesothelioma (-), with high specificity and sensitivity. Meanwhile, metastatic adenocarcinoma is a common factor causing malignant serous cavity effusion, while MOC-31 can label adenocarcinoma cells in hydrothorax and ascites, which plays a significant role in early screening and differential diagnosis of patients. Moreover, the high expression of MOC-31 is associated with poor prognosis, speculated that Ber-EP4 in tumors can interact with various cytokines to inhibit adhesion, maintain stemness, and mediate immune escape.
