Pan TRK is a monoclonal antibody that specifically labels the tropomyosin-related kinase (TRK) protein. TRK is a transmembrane receptor protein encoded by the neurotrophic receptor tyrosine kinase (NTRK) gene. TRK mainly exists in neurons, and its binding with ligands can promote cell proliferation and inhibit apoptosis. However, oncological research has found that NTRK gene can fuse with partner gene, and the protein encoded by them can continuously activate downstream signaling pathways without ligands, thereby stimulating the occurrence and development of tumors. NTRK gene fusion is commonly found in some rare tumors such as salivary gland or breast secretory carcinoma, congenital fibrosarcoma, and congenital mesoblastic nephroma. It can also be present at low frequency in common tumors such as thyroid cancer, colorectal cancer, and lung cancer. In clinical practice, Pan TRK has become an important biomarker for detecting NTRK gene fusion, and its positive expression suggests that patients can benefit from targeted therapy of TRK inhibitors such as Larotrectinib and Entrectinib.
