Transducin-like enhancer of split-1 (TLE1) is a transcription co-repressor that can interact with various DNA binding proteins to indirectly regulate gene expression, ensuring cell survival and inhibiting apoptosis. Synovial sarcoma (SS) is a malignant soft tissue sarcoma characterized by the t (X; 18) (p11.2; q11.2) chromosome translocation. Some research found that TLE1 is widely expressed in SS and is significantly associated with the translocation. This may be because TLE1 can bind to the fusion protein produced by the translocation and inhibit tumor cell apoptosis. Therefore, chromosome translocation detection combined with TLE1 immunohistochemistry has become the main diagnostic method for SS. In addition, TLE1 can be expressed in lung cancer, gastric cancer, pancreatic cancer, breast cancer, cervical cancer and other tumors, but it has different mechanisms in different tumors. For example, some scholars believe that TLE1 can reduce E-cadherin to promote epithelial-mesenchymal transition (EMT) of lung adenocarcinoma. But in pancreatic cancer, TLE1 can obstruct mitosis to inhibit tumor proliferation.
