Enhancer of zeste homolog 2 (EZH2) is a member of the polycomb group proteins, which can inhibit gene transcription by methylating the 27th lysine of histone H3 (H3K27), thereby participating in various life activities such as regulating cell survival, proliferation, migration, and senescence. The excessive production of EZH2 can be found in solid tumors such as liver cancer, gastric cancer, colorectal cancer, pancreatic cancer, prostate cancer, breast cancer, and is closely related to tumor development and drug resistance. Its tumor promoting effect involves multiple signaling pathways, such as the Wnt/β-catenin pathway that promotes proliferation, the JAK/STAT pathway that inhibits apoptosis, and the P13K/AKT pathway that mediates drug resistance. Meanwhile, the negative regulation of EZH2 on natural killer cells and the positive regulation of EZH2 on regulatory T cells play important roles in tumor immune escape. In addition, EZH2 is highly expressed in some lymphoid and hematopoietic system tumors such as multiple myeloma, B cell lymphoma, and NK/T cell lymphoma, indicating poor prognosis of patients.
