T-cell lymphoma/leukemia 1 (TCL1) is a dimeric protein that mainly exists in immature lymphocytes during embryonic development, participating in their proliferation and differentiation. In clinical practice, TCL1 is mainly used in the research and diagnosis of lymphoma. For example, TCL1 is abnormally overexpressed in T-cell prolymphocytic leukemia (T-PLL), and it is closely related to the occurrence and development of tumors. This is because TCL1, as an activator of AKT kinase, can continuously stimulate P13K-AKT signaling pathway, thereby promoting cancer cell proliferation and inhibiting apoptosis. Meanwhile, the high expression of TCL1 is considered as an independent risk factor for poor prognosis of chronic lymphocytic leukemia, diffuse large B cell lymphoma, and other tumor patients. In addition, some research has found that TCL1 can increase the invasiveness of liver cancer and colorectal cancer cells. It can not only increase AKT kinase activity, but also disable the inhibitory effect of WTX on β-catenin, thereby activating the Wnt/β-catenin signaling pathway and promoting epithelial-mesenchymal transition (EMT).
