PU.1 is a member of the ETS transcription factor family, which can regulate the normal development of hematopoietic cells by binding to DNA elements rich in GCAA/T gene sequences. The high level of PU.1 determine the differentiation of stem cells into macrophages and neutrophils, while the low level facilitate the differentiation into B lymphocytes. Clinical research has found that PU.1 abnormal expression will lead to abnormal development of hematopoietic cells. PU.1 is absent in classical Hodgkin lymphoma but expressed in various tumors such as B cell lymphoma and nodular lymphocyte-predominant Hodgkin lymphoma (NLPHL), making it become an important differential biomarker. Meanwhile, decreased PU.1 expression is considered as a triggering factor for acute myeloid leukemia (AML), and targeting PU.1 treatment has become a new research direction. However, some scholars pointed out that highly expressed PU.1 may be involved in the occurrence and development of erythroleukemia and mixed lineage leukemia, so the mechanism of PU.1 in different tumors still needs further research.
