Melanoma, also known as HMB-45, is a monoclonal antibody specifically targeting melanoma cells, which can specifically recognize the premelanosome glycoprotein gp100 in melanoma cells. Under physiological conditions, HMB-45 is mainly expressed in melanocytes during fetal period, but quiescent melanocytes in adults are negative, which only expressed in junctional nevus, Spitz nevus, and congenital nevus and other cells. HMB-45 is an indispensable marker for the differential diagnosis between benign and malignant melanoma. It has high specificity but low sensitivity, so it is often used in combination with other markers such as S-100 and Melan-A. In addition, some research has found that HMB-45 is expressed in various tumors such as angiomyolipoma, lymphangioleiomyomatosis, and clear cell sarcoma. It may be because some tumor cells are abnormally differentiated from precursor cells of melanocytes, which makes HMB-45 become a characteristic diagnostic indicator.
