Myogenin is a myogenic regulatory factor (MRF) that participates in the entire process of skeletal muscle development along with MyoD, Myf5, and MRF4. Under normal conditions, MRF only exists in skeletal muscle cells, while Myogenin is the only member existing in all skeletal muscle cell lines and the core regulatory factor for muscle cells terminal differentiation. Therefore, Myogenin mainly serves as a highly specific and sensitive diagnostic biomarker for rhabdomyosarcoma (RMS) in clinical practice. It is often used in combination with MyoD1 for the differential diagnosis of RMS (+) and leiomyosarcoma (-), RMS (+) and other small-round cell malignant tumors (-). Meanwhile, its expression results are correlated with the degree of tumor differentiation, which have guiding significance for prognosis evaluation. In addition, Myogenin is diffusely strongly expressed in alveolar RMS and unevenly lowly expressed in embryonal RMS, which makes it become one of the pathological classification reference indicators for RMS.
