Is there a standard counterstain time for hematoxylin?

At room temperature, counterstain for 8–10 s; adjust for section thickness and reagent strength. Too light reduces nuclear contrast; too dark masks positive signal. Follow with differentiation and bluing solutions per your post-staining workflow.
