[Intended Use]

This reagent is suitable for formalin-fixed, paraffin-embedded human tissue sections immunohistochemical staining,for counterstaining after the experimental procedure.

Hematoxylin(hematoxylin); potassium aluminum sulfate dodecahydrate;sodium iodate; composed of anhydrous citric acid, etc.

Store the reagent at room temperature in the dark.The validity period is shown on the outer packaging.

[Staining Principle]

Hematoxylin is a basic dye, that mainly stains chromatin in the cell nucleus purple-blue. Chromatin in the cell nucleus is mainly deoxyribonucleic acid (DNA)(DNA) ,DNA In the double helix structure of DNA, ,the phosphate groups on the two strands face outward, ,carry a negative charge, ,and are acidic, ,so they readily bind with positively charged hematoxylin basic dye by ionic or hydrogen bonds to be stained..

[Sample Requirements]

Fresh biopsy or surgical specimens are fixed with 10% neutral formalin solution for 6-24 hours, and processed into wax blocks by sampling, dehydration and paraffin embedding according to technical specifications. Wax blocks are valid for 5 years when stored at room temperature.

Section the specimen wax blocks to a thickness of 3-5μm, adhere them to anti-drop slides, and bake the slides in a constant temperature incubator at 58-60℃ for 1 hour for staining. Unused blank slides should be stored at room temperature (15-25℃) (preferably with anti-oxidation treatment). To well reproduce antigen distribution in tissue sections, immunohistochemical staining is recommended within 7 days.

[Staining Method]

After routine deparaffinization and hydration of tissue sections,rinse with tap water; ; or after immunohistochemical staining DAB after chromogenic development; ; stain tissue sections directly with this product ,for 3~5 minutes ((adjust according to actual conditions and needs)),After staining, rinse with tap water for 1 minutes , ammonia water (0.25%) or PBS(pH7.4)return blue 30 seconds, , then continue according to your respective operating procedure..

[Precautions]

1) This product is for research use only, not for other purposes.

2) Laboratory management should follow standard management specifications. Personnel must receive basic and professional training, wear lab coats and disposable gloves during experiments.

3) Take appropriate protective measures to avoid contact of the reagent with skin and eyes.

4) Disposal of toxic and hazardous reagents should follow the appropriate procedures.

5) When rinsing slides, do not flush directly onto the tissue to avoid tissue rupture or detachment.

6) This product slowly oxidizes when exposed to air; close the cap promptly after use to prevent oxidation.

7) This reagent can be reused until staining quality is unsatisfactory, then replace with fresh staining solution;

8) Use this reagent within the validity period.
